Week 5 of the 2026 NFL season concludes with an exciting interconference showdown as the Buffalo Bills travel to SoFi Stadium to take on the Los Angeles Rams on Monday Night Football at 8:15 p.m. ET on ESPN and ABC.

Buffalo enters the matchup at 3-1 after suffering its first loss of the season against the New England Patriots in Week 4. Los Angeles is 2-2 after rallying for a 24-20 victory over the Philadelphia Eagles.

Can Josh Allen and the Bills get back on track against a tough Los Angeles defense, or will Sean McVay's offense capitalize on Buffalo's defensive issues and secure another important home victory?

Below, we'll break down our Bills vs. Rams prediction and picks for Monday Night Football, including the spread and key player picks for PrizePicks.

Buffalo's offense has been one of the NFL's most efficient units through the first four weeks, ranking second in EPA per play (+0.092) and owning a 46.7% offensive success rate.

Most of that success hinges on QB Josh Allen, but RB James Cook has also been a major part of that production. Cook gives Buffalo an explosive rushing threat that can help take pressure off Allen, and he's running behind a line that's been one of the best at paving the way.

Buffalo's defensive efficiency is a major concern. The Bills are 27th in EPA/play (+0.033) on that end.

The Bills will miss two key contributors on Monday night, as DT Ed Oliver (knee) and WR Joshua Palmer (shoulder) have both been ruled out. Oliver's absence is especially significant against a dangerous, multifaceted Rams offense.

WR DJ Moore (shoulder), CB Dee Alford (ankle), and DL Landon Jackson (knee) are all questionable. CB Christian Benford is expected back in the secondary after missing Week 4, which would help against Matthew Stafford and Los Angeles' passing attack.

The Rams have been fantastic defensively, ranking third in EPA/play at -0.062 and allowing a 42.2% success rate.

Offensively, they've been less consistent but I have no doubt this offense will finish the season as one of the best in the NFL given the boatload of talent and the genius of McVay.

WR Puka Nacua returned from injury last week against Philly and didn’t miss a beat, finishing with 9 catches for 125 yards and a rushing TD. Speaking of injuries, the only real one to note on the Rams’ side is DT Aaron Donald, but he will suit up tonight.

Bills vs. Rams Prediction on Spread for Monday Night Football – 10/12/26

Here are the key stats and predictions behind our Bills vs. Rams pick against the spread for Monday night.

Unless otherwise noted, data used in this article is from SumerSports, PFF, and Pro Football Reference.

  • Allen is 23-16-2 against the spread (ATS) as an underdog.
  • Cook has been especially dangerous, trailing only Panthers RB Chuba Hubbard in explosive play rate.
  • Los Angeles frequently opts for light defensive boxes, sitting among the top five teams in usage. That approach could spell disaster against a run game as prolific as that of the Bills.
  • Buffalo's defense is second in pressure rate without blitzing, allowing the team to disrupt QBs while keeping extra defenders in coverage. Stafford loses nearly 0.6 EPA/play when pressured by four-man rushes.
  • McVay is 0-4 straight up in games officiated by Scott Novak, who's on tonight's matchup.
  • Under McVay, the Rams are just 3-9-1 ATS after winning their previous contest in which they trailed entering the fourth quarter.

Bills vs. Rams Picks, Player Stats for MNF on PrizePicks: 10/12/26

Here are some top Bills vs. Rams player picks to consider on PrizePicks for Monday night, including RB James Cook and WR Puka Nacua.

Bills RB James Cook vs. Rams: 75.5 Rush Yards

  • Cook averages 105.3 rushing YPG, and his workload is never a concern. He handles 94.6% of Buffalo’s RB carries, the highest share in the NFL.
  • He also has an 11.4% breakaway rush rate (first) and has failed to gain yardage on a league-low 5.7% of his runs.
  • PFF ranks Buffalo first in perfectly-blocked runs and yards before contact (2.36 per carry), creating opportunities for Cook to get into the open field.
  • The Rams’ defense has been a bit of a run-funnel thus far, ranking second in EPA/pass (-0.22) but 15th in EPA/run (-0.09).
Add James Cook + Bills to win to your PrizePicks lineup now.

Rams WR Puka Nacua vs. Bills: 94.5 Rec Yards

  • I love this spot for Nacua again vs. a Bills pass defense that ranks 26th in EPA/dropback (0.11).
  • Nacua's 12 targets against the Eagles suggest Stafford is comfortable going right back to him as the unquestioned alpha of the WR room.
  • Buffalo's defense ranks 30th in success rate allowed (48.16%).
  • The volume is shaping up to be plentiful tonight for Nacua, as the Bills allow 36.8 passing attempts per game (third most).
  • Nacua’s 26.6% target share sits 10th among all WRs, and he's playing in a game tied for the highest projected total of any Week 5 contest (54.5).
Add Puka Nacua + Rams to cover the spread to your PrizePicks lineup now.
